Overview
Triptorelin is a synthetic GnRH agonist (decapeptide analog) with significantly greater potency and longer half-life than endogenous GnRH. Continuous administration produces initial HPG axis stimulation followed by desensitization and suppression of gonadotropin release, a phenomenon exploited in both research and clinical applications.
Mechanism of Action
Triptorelin is a superagonist at GnRH receptors. Initial administration causes a "flare" of LH and FSH release (and consequent testosterone/estrogen surge). Continuous administration then downregulates GnRH receptors, suppressing pituitary gonadotropin secretion and reducing gonadal hormone levels to castrate/postmenopausal levels.
