Overview
Thymulin (Serum Thymic Factor, FTS) is a nonapeptide (9-amino acid) thymic hormone that requires zinc as a cofactor for biological activity. It is one of the key hormones produced by thymic epithelial cells and plays an essential role in T-cell maturation, differentiation, and immune system regulation.
Mechanism of Action
Thymulin binds to specific receptors on T-cell precursors, promoting their differentiation into mature functional T-cells. The zinc-thymulin complex is the active form; zinc binding induces the conformational change necessary for receptor recognition. Thymulin levels decline with thymic involution during aging, contributing to age-related immune decline.
Research Context
Thymulin levels in circulation decrease dramatically after puberty as the thymus involutes. This decline correlates with reduced T-cell function and increased susceptibility to infections and autoimmune conditions in aging. Zinc supplementation can partially restore thymulin activity.
