Overview
Thymosin Alpha-1 (Ta1) is a 28-amino acid peptide originally isolated from thymic tissue that plays a central role in immune system regulation. It is one of the most well-studied immunomodulatory peptides, with regulatory approval in over 30 countries for conditions including hepatitis B, hepatitis C, and immune deficiency. Its brand name pharmaceutical form is Zadaxin.
Mechanism of Action
Thymosin Alpha-1 activates toll-like receptors (TLR2, TLR9) on dendritic cells, promoting their maturation and antigen presentation. It enhances T-cell, NK cell, and dendritic cell function. It stimulates interferon-alpha and -gamma production, promoting antiviral and anti-tumor immunity. Ta1 also inhibits thymocyte apoptosis and supports T-cell differentiation.
Research Context
Clinical trials and clinical use data span viral hepatitis, HIV, cancer immunotherapy (as adjuvant), vaccination enhancement, and sepsis. Its safety profile across decades of use is well-established.
