Mazdutide (LY3305677 / IBI362) is a dual GLP-1/glucagon receptor agonist developed by Eli Lilly and licensed to Innovent Biologics. It combines GLP-1 receptor agonism for appetite control with glucagon receptor agonism for thermogenesis and hepatic fat oxidation, targeting the same receptor pair as two of retatrutide's three targets.
Mechanism of Action
GLP-1 receptor activation suppresses appetite and slows gastric emptying. Glucagon receptor activation promotes hepatic glycogenolysis, gluconeogenesis, and critically, hepatic fat oxidation and energy expenditure. The dual mechanism addresses both energy intake (appetite) and energy output (thermogenesis).

Clinical trials in China and globally have shown substantial weight loss with Mazdutide. It represents an alternative approach to the GIP/GLP-1 dual agonism of tirzepatide, substituting glucagon for GIP as the second receptor target.
