Overview
IGF-1 DES (Des(1-3)IGF-1) is a truncated variant of IGF-1 missing the first three N-terminal amino acids (Gly-Pro-Glu). This modification eliminates binding to most IGF-binding proteins, making DES-IGF-1 approximately 10x more potent than full-length IGF-1 at the receptor level due to its increased free fraction.
Mechanism of Action
IGF-1 DES activates the IGF-1 receptor with similar affinity to native IGF-1 but with dramatically reduced IGFBP sequestration. This means a much higher proportion of administered peptide reaches and activates IGF-1 receptors. The result is enhanced activation of PI3K/Akt and MAPK pathways driving protein synthesis and cell proliferation.
