Overview
Humanin is a 24-amino acid mitochondria-derived peptide (MDP) encoded by the 16S rRNA gene of mitochondrial DNA. Discovered in 2001 in surviving neurons of Alzheimer's disease patients, it was the first identified MDP and has since been recognized as a key player in cellular stress resistance, metabolic regulation, and aging.
Mechanism of Action
Humanin activates the STAT3 signaling pathway via a trimeric receptor complex (CNTFR/WSX-1/gp130). It also binds to IGFBP-3, competing with IGF-1 for binding and modulating IGF-1 bioavailability. Intracellularly, it interacts with BAX to prevent apoptosis and protects mitochondrial membrane integrity under stress conditions.
