Overview
Gonadorelin is a synthetic version of endogenous gonadotropin-releasing hormone (GnRH), a 10-amino acid peptide produced by the hypothalamus. It is the master hormonal signal that initiates the entire reproductive cascade by stimulating the anterior pituitary to release LH and FSH.
Mechanism of Action
Gonadorelin binds to GnRH receptors on pituitary gonadotroph cells, stimulating the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Pulsatile administration mimics physiological GnRH secretion and maintains gonadotroph responsiveness. Continuous administration paradoxically downregulates the receptor (desensitization), suppressing LH/FSH release.
Research Context
Gonadorelin is used diagnostically to assess pituitary function and therapeutically to stimulate gonadal function. Its dual nature (stimulatory when pulsatile, suppressive when continuous) makes it a versatile research tool.
Key Research Areas
Considerations for Researchers
Pulsatile administration requires precise timing protocols. The peptide has a short half-life (2-4 minutes). Store at -20C. Fresh reconstitution recommended for each use.
