Overview

CJC-1295 with DAC (Drug Affinity Complex) is a synthetic GHRH analog with a maleimidopropionic acid modification that covalently binds to serum albumin in vivo. This DAC technology extends the half-life from minutes (native GHRH) to approximately 6-8 days, enabling sustained GH elevation from a single administration.

Mechanism of Action

CJC-1295 with DAC binds GHRH receptors on anterior pituitary somatotrophs, stimulating GH synthesis and secretion. The DAC modification creates a depot effect by binding to circulating albumin, resulting in sustained receptor stimulation and chronically elevated GH and IGF-1 levels. This produces a continuous elevation rather than the sharp pulses seen with non-DAC versions.

Research Context

Studies have shown that a single dose of CJC-1295 with DAC produces elevated GH levels for 6+ days and elevated IGF-1 for 9-11 days. While this provides research convenience, the continuous GH elevation is less physiological than pulsatile release patterns.
