Overview
CJC-1295 without DAC (also called Modified GRF 1-29) is a synthetic GHRH analog with four amino acid substitutions that improve metabolic stability compared to native GHRH. Without the Drug Affinity Complex, it has a half-life of approximately 30 minutes, producing acute GH pulses rather than sustained elevation.
Mechanism of Action
CJC-1295 no DAC binds to GHRH receptors on pituitary somatotroph cells, triggering a sharp pulse of growth hormone release that mimics the natural pulsatile GH secretion pattern. The four amino acid substitutions (at positions 2, 8, 15, 27) protect against enzymatic degradation by DPP-IV and other peptidases without fundamentally altering the pharmacodynamic profile.

The no-DAC version is generally preferred in research protocols that aim to study pulsatile GH release, as it better approximates physiological GH secretion patterns. It is commonly paired with Ipamorelin for synergistic effects.
