Overview
BDNF (Brain-Derived Neurotrophic Factor) is a 247-amino acid protein belonging to the neurotrophin family. It is the most abundant and widely distributed neurotrophin in the brain, playing critical roles in neuronal survival, differentiation, synaptic plasticity, and memory formation.
Mechanism of Action
BDNF binds to TrkB (tropomyosin receptor kinase B), activating three major signaling cascades: PI3K/Akt (cell survival), Ras/MAPK (neuronal differentiation), and PLCgamma (synaptic plasticity). These pathways collectively support long-term potentiation (LTP), the cellular basis of learning and memory.
Research Context
BDNF is central to the neurotrophin hypothesis of depression and cognitive decline. Low BDNF levels correlate with depression, anxiety, neurodegeneration, and age-related cognitive decline. Exercise, certain dietary patterns, and several peptides (Semax, Selank, Dihexa) increase endogenous BDNF.
