Overview

Melanotan-1 (Afamelanotide) is a synthetic linear analog of alpha-MSH with greater MC1R selectivity than Melanotan-2. This selectivity produces melanogenic effects (skin darkening) with significantly fewer off-target effects on sexual function and appetite compared to the broader-acting MT-2.

Mechanism of Action

Melanotan-1 selectively activates MC1R on melanocytes, stimulating eumelanin production. Eumelanin is the brown-black pigment that provides UV protection. The targeted MC1R activation avoids significant CNS effects (sexual arousal, appetite changes) associated with MC3R/MC4R/MC5R activation seen with MT-2.

Research Context

Afamelanotide has been approved in Europe (as Scenesse) for erythropoietic protoporphyria, a condition where patients cannot tolerate sunlight. Research continues into photoprotection, vitiligo, and skin cancer prevention applications.
