Overview

KPV is a C-terminal tripeptide fragment of alpha-melanocyte-stimulating hormone (alpha-MSH), consisting of the amino acids Lysine-Proline-Valine. Despite being only three amino acids long, it retains the potent anti-inflammatory activity of the parent hormone while lacking its melanogenic (skin-darkening) effects.

Mechanism of Action

KPV enters cells and translocates to the nucleus, where it inhibits NF-kB activation by preventing the phosphorylation and degradation of IkBalpha. NF-kB is a master transcription factor controlling the expression of pro-inflammatory cytokines including IL-1beta, IL-6, IL-8, and TNF-alpha. By blocking this pathway at the nuclear level, KPV produces broad anti-inflammatory effects.

Research Context

Research has focused on KPV's anti-inflammatory properties in gastrointestinal and dermatological models. Studies in colitis models demonstrate reduced mucosal inflammation and improved barrier function. Skin research shows reduced inflammatory cytokine production and improved wound healing.

Key Research Areas

NF-kB inhibition mechanisms and nuclear translocation
Inflammatory bowel disease and colitis models
Skin inflammation and dermatitis research
Gut barrier function and tight junction integrity
Oral bioavailability of the tripeptide
Comparison with full-length alpha-MSH

Considerations for Researchers

The small size (3 amino acids) contributes to stability and potential oral bioavailability. Available in injectable and oral forms. Some vendors offer KPV in capsules for gut-targeted research. Store at -20C lyophilized.
