DSIP (Delta Sleep-Inducing Peptide)

Overview

DSIP (Delta Sleep-Inducing Peptide) is a naturally occurring nonapeptide (Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated from rabbit brain in 1977. Named for its ability to induce delta (slow-wave) sleep in animal models, it has since been found to have broader neuroendocrine regulatory functions beyond sleep architecture.

Mechanism of Action

DSIP modulates multiple neurotransmitter systems including GABAergic, glutamatergic, and serotonergic pathways. It influences corticotropin-releasing hormone (CRH) secretion and hypothalamic-pituitary-adrenal (HPA) axis activity, contributing to stress modulation. Its sleep-promoting effects are thought to involve facilitation of the transition from wakefulness to slow-wave sleep rather than sedation.

Research Context

Research has demonstrated DSIP effects on sleep architecture, pain perception, stress hormone regulation, and withdrawal symptoms in substance dependence models. It does not produce the forced sedation of traditional sleep medications but rather promotes natural sleep onset and architecture.
